Storing Free-Floating Sections
Antigen Preserve (AP) Solution
All MultiBrain sections are placed into AP storage solutions during sectioning. AP maximizes the antigenicity of proteins while eliminating concerns about formalin-sensitive staining which may be applied to the sections.
AP Solution Composition:
• PBS mixed as directed by manufacturer pH 7.0 – pH 7.4 with dH2O
• As a 10x concentrate, 1 part of liquid will be mixed with 9 parts of dH2O
• E.g. 100 ml of concentrate, 900 ml of dH2O
• Ethylene glycol
• Polyvinyl pyrrolidone (PVP)
Use these amounts or others in the same proportions, duly recorded, to make the required total volume:
Component | 2000 ml | 3000 ml | 4000 ml | 5000 ml | 6000 ml |
---|---|---|---|---|---|
10x PBS Concentrate | 100ml | 150 ml | 200 ml | 250 ml | 300 ml |
dH2O | 900 ml | 1350 ml | 1800 ml | 2250 ml | 2700 ml |
